We claim that the IBIS MX96 (IBIS Technologies, Enschede, Netherlands) is the only instrument that meet our requirements. Our experiments are expected to include:

— Multiplex kinetic and steady-state characterization of binding interactions over a large range of immobilization levels from limited sample amount

— Multiplex epitope binning and epitope mapping

— Fundamental studies of binding within macromolecular complexes

— Multiplex screening of complex proteomic samples for identification of interactions partners

— Recovery of injected and captured sample for mass spectrometric analysis

— Fundamental studies of applications within SPR imaging

To be able to conduct these studies in high quality we need:

— Real-time SPR imaging with continuous scanning of SPR angle

— low sample consumption (<150 µl per sample) whilst being able to inject sample at non-diffusion limiting flow rates for >60 minutes to improve sensitivity

— large linear dymanic range, between ~ 1.33-1.37 RIU (40,000 RU). Baseline noise < 0.5 RU

— Possibility to elute from sensorarea and to recover injected sample

— Multiplexing capabilities of at least up to 96 interactions

— Dedicated software that supports kinetic and epitope binning analysis of the multiplex data

— Software that allows unattended analyses >24 h

Important features and benefits of the IBIS MX96 that we need and which differentiate this system from other commercially available offering include the combination of:

— Ultra low analyte consumption of 100 microliters that can be recovered,

— Unlimited interaction times enabled by back and forth mixing

— Valve-less injection of samples for improved stability, even with complex samples

— SPR imaging allows real-time 'visual' assessment of array quality and analysis with continuous scanning of SPR angle

— Largest dynamic and linear range enables full compensation of 'common mode' effects by referencing

— Multiplexing with 96-plex array fabrication (CFM) in a double print

— Local (individual) referencing for 96 reaction spots and calibration of the ROI sensitivity reveals unsurpassed quality on multiplexed data

— Data analysis in SPRintX software and an adapted version of Scrubber

— Dedicated epitope binning software for high-throughput data analysis
