The technology/platform/chemistry must meet the following demands: 1) It must be based on single-molecule sequencing and not demand any PCR or amplification of native molecules. 2) It must allow for rapid, real-time sequencing with maximum runtimes not exceeding 48 hours. 3) It must offer the potential for the generation of single sequencing reads in excess of 100Kb. 4) It must be usable for sequencing both DNA and RNA, and for the detection of methylation status. 5) It must offer a variety of sample preparation protocols including protocols suitable for sequencing DNA that take less than 15 minutes from DNA to first real-time sequence. Our decision to buy a sequencing bundle from this supplier is based on the fact that this unique technology enables the generation of very long reads (>100Kb) from single DNA molecules in real-time. Such reads are very well suited to our goals including de novo sequencing of large (>1Gb), fish and plant genomes.
