Short description

Automated computer-assisted DNA/RNA synthesizer
1) Automated computer-assisted DNA/RNA synthesizer (including all necessary parts required for the action in passage 2)
2) Capable for the automated solid phase synthesis of oligonucleotides by phosphoramidite chemistry, including an option for H-phosphonate chemistry. The synthesizer should be capable for the synthesis of oligonucleotides with a native phosphodiester backbone, full phosphorothioate backbone, and also with a phosphorothioate-native phosphodiester mixmer backbone .
3) The synthesizer should be equipped with an UV-/conductivity detector suitable for monitoring of the coupling efficiency/progress of the assembly (primarily detection of DMTr removal).
4) The synthesis cycles should be reprogrammable by the user . The program files should contain optimized standard programs for small scale synthesis (1-2 µmol), for the synthesis of both DNA and RNA with both native phosphodiester and phosphorothioate backbone, and for the synthesis of their hybrids (gapmers).
5) The synthesizer should contain at least 8 reagent vessels (12 optimum) for different building blocks, appropriate reagent vessels/lines for the deblocking (typically 3% dichloroacetic acid in dichloromethane or toluene), oxidation (typically I2 in a mixture of pyridine, water and THF), sulphurization (different options), activation (e.g. benzylthiotetrazole solution), and two lines for capping.
6) The synthesizer should be capable for parallel synthesis of different sequences, or at least to re-programmed sequence by sequence synthesis.
7) The equipment should be flexible for different scales, but the optimum being on 1-2 umol scale.
8) The total price should contain i) delivery of the equipment (all items included into the tender) to the Laboratory of Organic Chemistry and Chemical Biology, Department of Chemistry, University of Turku and ii) training to the equipment at the time of installation. The training should contain attendees with a complete overview of the equipment and its software, practical aspects of operation and covering basic maintenance.
9) Guarantee offered for the equipment (In minimum 12 months, including all costs involved instrument repairs, parts, labour expenses, travel expenses and plausible expenses, if the equipment should be returned to the supplier).
10) During and after guarantee period capability to provide on-site service by competent personnel is mandatory within 48h (excluding saturday, sunday), unless other means (f.ex email, phone instructions) are not enough to resolve the problem. As an addition to JYSE 2014 Supplies terms failing to follow timeline results in penalty fee 0,25%xoffered total price of a tender for each starting late 24 hour. Tenderer shall not be allowed to create any pricing formula which aims nullifiyng the effect of penalty fee term.
